So, it's the market price of the stock at close on Friday that matters?
If it's $300 all day, then plummets $50 at 3:59 and closes at $50, then that option i used in my scenario just cost me $6650?

The option prices don't change when the market is closed.

Options expire at the end of the day on the expiration day

Yes. And most places will automatically exercise options that expire in the money.
