No, not sure when the first was, but it happens fairly frequently, 2003, 2004 etc

First one listed there is 2003 Byron Leftwich beating the Bucs, but I'd also strongly guess there where some in the century prior
Unless you mean that Max Hall was the first rookie in that specific season to beat the Saints (who were the defending Super Bowl champs), doing it a few weeks before Colt McCoy also beat the defending Super Bowl champs in that same season
